Surface plasmon resonance (SPR) technology was developed for BIACORE X biosensor from Biacore AB Company. SPR is an optical system used for detecting real time biomolecular interaction when a sample was injected into the instrument and interacted with a ligand which was covalently attaching to the sensor surface. Cell transudation promote carcinogen protein Ras action reaction by recognition of SH2 domain of Grb2 protein by the active receptor of growth factor. SH2 domain regulates specific protein-protein interaction through linkage with specifically phosphoryl tyrosine peptides, SH2 domain of Grb2 recognizes pYXNX for interacting with EGFR protein. We have analyzed protein-protein interaction by SPR technology and established SPR-based method for screening peptides which inhibited the interaction of Grb2 SH2 domain. The p-sensor 2000 designed by ANT is based on the principle of piezoelectric biosensor. It is made of three portions including electronic oscillation circuit, frequency counter, and piezoelectric quartz of fixed biosensor molecule (p-chip). Piezoelectric quartz crystal consist of quartz crystal slab with a layer of gold electrode on each side. It is the signal conversion component of piezoelectric sensor chip and can convert the result sensed by the sensor molecule into electronic signal and being amplified. The p-sensor 2000 does not need other labeled material such as enzyme and radioactivity to obtain the dynamics of interaction between molecules. It has the advantages of high sensitivity, fast response, and instant inspection. All kinds of inspection such as antibody/antigen, enzyme / substrate, hormone / receptor, and nucleic acid / nucleic acid that based on biotic affinity reactions can be measured by fixing different sensor molecule on the piezoelectric crystal gold electrode for the different analyte.The purpose is to transfer the coating ligand methods and analysis of SPR technology to QCM technology?Ausing our already established Grb2 SH2 research programs. And compare between two biochip technology, to further develop the specifically, stability and fast biochip analysis.
