    题名: 洋蔥、芸香苷之代謝動力學及芸香苷對急性腎衰竭之影響;Metabolic Pharmacokinetics of Onion, Rutin and Effect of Rutin on Acute Renal Failure

    摘要: 洋蔥 (Allium cepa L. 之鱗莖) 之多酚含量居蔬果之冠,主要多酚為槲皮素配醣體 (quercetin glycosides),以洋蔥皮含量最高。大鼠口服洋蔥皮及洋蔥湯後經藥物動力學模式,於特定時間由心臟採血,血清以高效液相層析法定量。結果顯示血中無 quercetin 之原型,主要以結合態代謝物 quercetin sulfates 存在。洋蔥湯中 quercetin glycosides 之吸收明顯較洋蔥皮快,而洋蔥皮可視為緩釋劑型。
    胺甲葉酸 (Methotrexate,MTX) 為治療指數狹窄的抗癌藥及免疫抑制劑。大鼠採平行設計,探討分別口服洋蔥皮及洋蔥湯對MTX動力學之影響。結果顯示併服洋蔥皮對 MTX 之血藥面積顯著提高了 140 %,平均滯留時間顯著提高了 90 %,大鼠死亡率 100 % (5/5)。併服洋蔥湯對 MTX 血峰濃度顯著提高了 100 %,血藥面積顯著提高了 99 %,但對平均滯留時間未達顯著影響,大鼠全數存活。此動力學交互作用結果顯示,洋蔥皮雖然是多酚的豐富來源,但與 MTX 併服卻有增加其毒性的風險。
    大鼠肌肉注射 50 % glycerol 誘發急性腎臟衰竭模式中,藉由監測血中 BUN、creatinine 評估 rutin 對腎臟功能的影響。結果顯示口服500 mg/kg rutin 顯著降低 BUN、creatinine,具減緩急性腎臟衰竭進展之功效。

    Onion (bulbs of Allium cepa L.) is the richest source of polyphenols among vegetables and fruits. The major polyphenols in onion are quercetin glycosides and onion skin has the highest content. This study investigated the pharmacokinetics of onion skin and onion soup in rats. Our results indicated that after oral administration of onion skin and onion soup, no free form of quercetin was detected, whereas quercetin sulfates were found predominantly in the blood. Onion soup showed faster absorption of quercetin glycosides than onion skin, which is like a sustained-released dosage from.
    Methotrexate (MTX), a dicarboxylic acid, is an important immunosuppressant with narrow therapeutic index. Rats were orally given MTX with and without onion skin and onion soup. The results showed that coadministration of onion skin significantly increased AUC0-t of MTX by 140 % and increased MRT by 90 %. In addition, unexpectedly, coadministration with onion skin resulted in a mortality of 5/5 (100%). Coadministration of onion soup significantly increased the AUC0-t of MTX by 99 %, but didn’t affect MRT and all rats survived well throughout the study.
    Intramuscular administration of 50 % glycerol induced acute renal failure of rats. Our results showed that oral administration of 500 mg/kg of rutin significantly reduced serum creatinine and BUN, indicating that rutin exerted kidney protection effect.
