    摘要: 二甲基甲醯胺(DMF)是已知的人類肝毒性化學物,對動物造成惡性腫瘤發生,但是其對人類基因毒性之效應目前還不清楚。本研究目的是利用彗星試驗(comet assay)來評估合成皮工廠作業員工之空氣中二甲基甲醯胺個人曝露量與體內DNA損傷之間的關連性,並且評估勞工DMF曝露量對其健康之影響。

    本研究對象是中部某皮革工廠之67名勞工,配合勞工健康檢查前隨機選取30名勞工測量個人空氣DMF八小時日時量平均濃度,同時抽取每名研究對象靜脈血檢測DNA損傷程度及生物指標。問卷資料的收集是以獨立個人訪視的方式進行,問卷的資料包括個人基本資料、過去的病史、工作史、抽菸、飲酒及疾病病史。

    研究對象依工作區分為暴露組37人與對照組30人。暴露組平均年齡為37.1歲,對照組為36.5歲,兩組在統計上沒有差異。暴露組有26人(70.3%)抽菸,平均抽菸量為12.8包年,對照組有6人(20.0%)抽菸,平均抽菸量為6.9包年,統計達到顯著差異。暴露組(n=27)的麩氨酸草酸轉化脢(AST)為30.4 10.1 IU/L、對照組(n=30)為23.9 8.1 IU/L在統計上有明顯的差異(p=0.009)。暴露組的麩氨丙酮酸轉化脢(ALT)為40.2 20.2 IU/L、對照組為26.1 15.4 IU/L在統計上有明顯的差異(p=0.004)。暴露組的肌酸酐為1.0 0.1 mg/dL、對照組為0.8 0.2mg/dl在統計上有顯著意義(p<0.001)。個人作業環境測定依照濃度分為高(8.3ppm)、中(3.0ppm)、低(0.3ppm)暴露量,DNA損傷程度高暴露族群與中暴露族群各為186.2 45.0與143.8 40.8在統計學上有顯著差異(p=0.028)。高暴露組186.2 45.0與低暴露組144.1 38.1也有差異性(p=0.043)。

    本研究利用彗星試驗探討評估二甲基甲醯胺暴露勞工之DNA損傷程度,結論發現暴露高濃度DMF之勞工比暴露於低濃度之勞工DNA受損的程度在統計學上有顯著的差異。暴露於DMF之勞工其肝功能有顯著性的差異。

    N,N-dimethylformamide(DMF) is a well-known hepatotoxic chemical, and has been shown to cause malignancy in animals. However its genotoxicity in humans is unclear. The purposes of this study examine associations of airborne DMF levels and DNA damage using the comet assay, and them health stating in synthetic leather workers. A questionnaire was administered to obtain detailed occupational, smoking, alcohol consumption, and medication histories.
    Thirty-seven works of expose group and thirty works of control group were divided. Mean age of expose group was 37.1 years old and controls was 36.9 years old, they was notsignificant. There were 26 smokers (70.3%) exposd group,and mean-pack years was12.8. There were 6 smokers (20.0%) in controls ,mean in controls pack-years was 6.9. AST levels in exposd group (30.4 10.1【n=27】and in controls 23.9 8.1【n=30】) were significantly. ALT levels in exposd group (40.2 20.2 【n=27】and in controls 26.1 15.4【n=30】) were significantly. Creatinine levels in exposd group (1.0 0.1【n=27】 and in controls 0.8 0.2【n=30】) were significantly.
    The DNA damage level for works in the high DMF (8.3ppm) and middle DMF (3.0ppm), 186.2 45.0 and 143.8 40.8,and showed a significant difference. The DNA damage level for works in the high DMF (8.3ppm) and low DMF (0.3ppm), 186.2 45.0 and 144.1 38.1,and also showed a difference.
    This study using the comet assay to assess the level of DNA damage in DMF exposed workers. We found that workers who exposed to high concentration DMF had great DNA damage than those exposed to low level of DMF. Exposure DMF workers also has more hepatotoxic effect than controls.
