    Title: 台灣產大頭艾納香之鎮痛及抗發炎研究
    Studies on the analgesic and anti-inflammatory activities of Blumea riparia (Blume) DC. var. megacephala Randeria

    Abstract: 大頭艾納香為菊科艾納香屬植物大頭艾納香(Blumea riparia (Blume) DC. var. megacephala Randeria)的莖、葉部分,為台灣民間常見之藥用植物。本論文以體外試驗及動物實驗探討大頭艾納香甲醇萃取物之自由基清除能力、鎮痛及抗發炎作用之分子機轉:
    BRMeOH具有清除DPPH自由基能力、Trolox當量的總抗氧化能力測定、還原力測定及Ferric reducing/antioxidant powder(FRAP),顯示其具有良好的抗氧化能力。
    鎮痛實驗方面,BRMeOH (0.1,0.5 g/kg)可明顯減少由醋酸扭體實驗的小鼠疼痛扭動次數,證實BRMeOH具有痛效果。
    在抗發炎方面,BRMeOH (0.5 g/kg)可明顯抑制由λ-carrageenan所誘導的小鼠足蹠發炎腫脹,其足蹠組織中cytokines (IL-1β、IL-6、TNF-α)以及NO表現量明顯受到抑制。綜合上述結果顯示,BRMeOH (0.5 g/kg)具鎮痛、抗發炎之作用。其鎮痛機轉為減少發炎反應而達到;其抗發炎的機制可能由以下兩種機轉:(1)抑制cytokines (IL-1β、IL-6、TNF-α)的含量,進而抑制NO的生成,減少自由基的傷害;(2)抑制cytokines (IL-1β、IL-6、TNF-α)的濃度而達到抗發炎的作用。
    Blumea riparia (Blume) DC. var. megacephala Randeria is the common folk medicinal plant in Taiwan, which belongs to Asteraceae family. In this thesis, in vitro tests and animal experiments were exployed to the radical scavenging capacity of methanol extract of Blumea riparia.The molecular mechanism of the analgesic and anti-inflammatory effects were also evaluated.
    DPPH scavenging activity test, Trolox equivalent antioxidant capacity determination, determination of reducing power and Ferric reducing / antioxidant powder (FRAP) indicated that the methanol extract of Blumea riparia had strong antioxidant capacities.
    The methanol extract (0.1,0.5 g/ kg) of Blumea riparia can significantly inhibit the writhing response in acetic acid writhing test.
    The methanol extract (0.5 g/ kg) of Blumea riparia can significantly inhibit the λ-carrageenan-induced mice paw swelling and inflammation. It also inhibited cell infiltration and increased neutrophils. The plantar tissue of cytokines (IL-1β, IL-6, TNF-alpha) and NO expression were significantly suppressed.
    The results showed that the methanol extract (0.5 g/ kg) of Blumea riparia exhibited analgesic, anti-inflammation effect. The analgesic effect was achieved due to reduction of the inflammation. The anti-inflammatory effect may be due to the following two mechanisms: (1) inhibition of cytokines (IL-1β, IL-6, TNF-α) content, thereby inhibiting the formation of NO reduce free radical damage; (2) inhibition of the concentration of cytokines (IL-1β, IL-6, TNF-α),concentration to achieve the anti-inflammatory role.
