題名: | 川芎對於streptozotocin 誘發高血糖大鼠學習障礙暨肝腎功能之影響 The effect of Ligusticum chuanxiong on learning disabilities and the function of liver and kidney in streptozotocin-induced hyperglycemic rats |

摘要: | 本研究探討川芎對streptozotocin(STZ)誘導之高血糖鼠在學習障礙,及肝、腎功能的影響,並且測定腦、肝、腎及胰臟的抗氧化酵素活性,以了解川芎對高血糖鼠改善學習障礙與肝、腎等功能的機轉。
本實驗將大鼠分成七組:( 1 ) 對照組 ( 2 ) STZ誘導高血糖組 ( 3 ) 川芎低劑量治療組 ( 4 ) 川芎高劑量治療組 ( 5 ) 川芎?低劑量治療組 ( 6 ) 川芎?高劑量治療組 ( 7 ) 阿魏酸治療組。結果發現,川芎粗抽物對STZ誘導之高血糖鼠具降低血糖之作用,而STZ誘導之高血糖鼠會造成肝臟方面的損傷,在血液中γ- GT、AST以及ALT的含量會明顯增加。
STZ誘導高血糖組在plus maze、water maze以及probe test都會造成學習記憶障礙的現象,而在給予川芎粗抽物、高劑量川芎?及阿魏酸的治療組,對STZ誘導高血糖鼠所誘發之學習記憶障礙有改善的效果。
另外,STZ誘導之高血糖鼠在海馬的AChE活性也有明顯降低,在total GSH、total SOD以及Mn-SOD的含量都有降低,而在海馬、肝臟與腎臟中的MDA有升高的情形。
綜合以上各點,STZ誘導之高血糖鼠在學習記憶方面以及肝功能可能都有所損傷情況,而川芎粗抽物與阿魏酸可能藉由回復AChE的活性,在十字迷宮和水迷宮可看出對於高血糖鼠是有學習記憶改善的趨勢,在肝功能部份,川芎與川芎?可能藉由降低MDA而去改善或修復肝臟機能。
This study investigated the effect of Chuanxiong for streptozotocin (STZ)-induced hyperglycemic rats in learning and memory, liver and kidney function, and determination in the brain and other organs of the antioxidant enzymes, then we could find Chuanxiong for hyperglycemia rat have the ability to improve memory and learning as well as other functions.
In this study, rats were divided into seven groups (1) control (2) STZ-induced hyperglycemia (3) low-dose of Chuanxiong crude extract (4) high-dose of Chuanxiong crude extract (5) low-dose TMP (6) high-dose TMP (7) ferulic acid. The experimental results showed that the crude extract of Chuanxiong for STZ-induced hyperglycemia rats have the effect of reduce blood glucose, and liver function in STZ-induced rat would cause damage in the blood γ-GT, GOT and GPT levels significantly increases. In renal function, STZ-induced hyperglycemia rats is similar to the control group.
STZ-induced hyperglycemic rats in learning, memory and liver function may have the injury situation, and Chuanxiong crude extract and ferulic acid may reply by AChE activity in the plus-maze and water maze.We could seen for that the hyperglycemia rats improved learning and memory. In liver function, Chuanxiong and TMP decreased MDA to improve or repair the liver function.
The content of STZ-induced hyperglycemia in the hippocampus AChE was significantly lower, in total GSH, total SOD and Mn-SOD would reduced the content , while in the hippocampus, liver and kidney MDA level had elevated.
It was found the STZ-induced hyperglycemia group in plus maze, water maze and probe have learning and memory impairment problems, and then give Chuanxiong crude extract, high doses of TMP and ferulic acid for STZ-induced hyperglycemia rat.Their learning and memory impairment has improved. |
