    摘要

    三葉茀蕨為水龍骨科植物三葉茀蕨(Crypsinus hastatus(Thunb.)Copel.)的全草。民間用於祛風解熱、消腫、結核、肝炎、痢疾、解毒等,為常用民間藥,台灣分布於全省山地潮濕林下岩石上。
    本研究以三葉茀蕨甲醇萃取物,經各有機溶媒分劃得正己烷層、氯仿層、乙酸乙酯層、正丁醇層及水層,以DPPH自由基清除能力試驗方法,評估三葉茀蕨甲醇萃取物及各分層的抗氧化活性;並以甲醇萃取物對ICR系雄性鼷鼠檢測其急性毒性試驗。當因檢品為2 mg/mL時,正丁醇層之抗氧化能力為95.42%,結果顯示三葉茀蕨之DPPH自由基清除能力以正丁醇層為最佳,其次依序為乙酸乙酯層,氯仿層。而急性毒性試驗方面,三葉茀蕨甲醇萃取物其LD50>10 g/kg,可認定三葉茀蕨是安全的,但真正臨床劑量仍需進一步探討。
    在成分研究方面,由三葉茀蕨經分離純化及結構鑑定後,分離出的成分有coumarin (H-1)、β-sitosterol (H-2)、20-hydroxyecdysone (E-1)、β-ecdysone (E-2)、naringin (E-3)等成分。而在coumarin定量分析結果顯示每克三葉茀蕨甲醇萃取物中coumarin含量約為8.03 μg,換算得到每克三葉茀蕨甲醇萃取物之含量百分比為0.0803%。而在naringin定量分析結果顯示每克三葉茀蕨甲醇萃取物中naringin含量約為0.56 mg,換算得到每克三葉茀蕨甲醇萃取物之含量百分比為5.6%。

    Abstract

    Crypsinus hastatus(Thunb.)Copel. is a native plant of Taiwan belonging to Polypodiaceae family. It is a common folk medicine used for dispelling colds, detumescence, tubercle bacillus, hepatitis, dysentery and detoxication, etc. It grows under the rock at mountain areas in Taiwan.
    The methanol extract of whole plant C. hastatus was fractionated into n-hexane, chloroform, ethyl acetate, n-butanol and aqueous fractions. The antioxidant activity of methanol extract and various fractions were evaluated by DPPH free radical scavenging activity model. The acute toxicity of methanol extract in rats was also evaluated. The result revealed that the n-butanol fraction at two milligram per milliliter exhibited 95% antioxidant activity was ninty-five percent. It exhibited better antioxidant potency than ethyl acetate and chloroform fractions in DPPH free radical scavenging activity. In the acute toxicity test the LD50 of the methanol extract of C. hastatus was larger than 10g/kg. It is considered relatively safe.
    In the phytochemical study, compounds isolated from Crypsinus hastatus were identified as coumarin (H-1), β-sitosterol (H-2), 20-hydroxyecdysone (E-1)、β-ecdysone (E-2) and naringin (E-3). The coumarin and naringin contants in the methanol extract of C. hastatus were also analyzed as 0.0803% and 5.6% separately.
