    摘要: 癌症一直以?是我國十大死亡原因的首位,並為全球人?所共同面?的嚴重疾病,近 ???癌患者?齡層有逐?下?之趨勢,癌症防治就成為近代醫藥研究最嚴峻的挑戰 之一。因此,世界各先進國家均投入相當的人?、物?,積極尋求突破。本計畫的目 標在於整合三大頂尖癌症中心,包括台大醫學院、中國醫藥大學暨醫?體系及美國安 德森癌症中心,進?積極且多元化的多邊合作關係,以期建?世界級跨國合作的癌症 研究中心。透過發現癌症新標記 (Biomarkers)、研究分子致病機轉、探討訊息傳遞網 ?及代謝體(Metabolomics) 的分子機制,進而發展新穎的標靶治?藥物以?為未?新 穎的?床試驗鋪?。此跨國合作的癌症研究中心之終極目標,將提升台灣的癌症研究 至國際一?水準,並預期對於早期癌症的偵測?及癌症病患的存活?具有具體的貢 獻。本計畫整合?台大醫學院和中國醫藥大學暨醫?體系的癌症研究團隊,以達成「建 ?世界級一?癌症研究中心」為目標。因此與安德森癌症中心成?的跨國頂尖研究中 心,其研究?域規劃係依據既有雄厚堅實的研究基礎,進而將基礎研究成果協助發展 新穎治癌藥物和新診斷技術之?床醫?系統,使得此跨國研究中心具備足夠的潛?成 為世界級頂尖的癌症研究中心。在“bench to bedside”的研究方面,以探討癌症的分子 機制為主,研究?域囊括?基因體平台、代謝體平台、訊息傳遞網?平台、藥物設計 生物資訊平台、藥物篩選平台、抗癌藥物之動物模型、分子??病學平台、幹癌細胞 平台、生物標本庫等,期許能協助發展新穎的標靶基因、治癌藥物與診斷技術。在 “bedside to bench”的研究方面,以雙方「癌症卓越研究中心」為主,以解決腫瘤患者 ?床治?所遭遇的問題為宗旨,如抗藥性的產生、癌症轉移、癌症?發、治?導致的 嚴重併發症等,以能發展個人化醫?自許。 計畫總目標: 目標一: 找出腫瘤形成、遠端轉移及抗藥性的新穎分子機制,尤其是具有東西方種族 差?的癌症種?。 目標二: 找出具有藥物研發潛?的標靶分子與?徑。 目標三: 發展腫瘤偵測指標及有效治?策?,改善癌症病患預後情況。

    Cancer is one of the leading causes of mortality in the world. Many interventions have been developed to circumvent issues that were encountered in diagnosis, treatment and prevention, yet there is still the need for improvement. Thus, the goal of the project of ‘Top International Research Center’ is to combine existing strengths of both cancer centers in Taiwan, College of Medicine, National Taiwan University and China Medical University & Hospital, and MD Anderson Cancer Center, USA and to improve diagnosis, treatment and prevention for cancer patients in Taiwan. The research will be focused on discovering new biomarkers, molecular mechanisms of pathogenesis and metabolomics, signal transduction pathways of cancer cells and the role of cancer stem cells with the combination of drug discovery and development of novel targeted therapy. This project combines the leading cancer research teams of NTUCM and CMU&H, to achieve the goal of ‘establishing a top cancer research center in the world’. As the current ‘Top International Research Center’ proposed by NTUCM, CMU&H and MD Anderson already has a strong basic research foundation, the main objective of this center is to apply and translate basic research into new drug development and new diagnostic techniques. This center will have a great potential to become a top cancer research center in Asia. One research direction for "bench to bedside", is to explore the molecular mechanisms of cancer; the research fields include genomics, metabolomics, signal transduction pathway, drug design and screening, animal models for anti-cancer drug, molecular epidemiology and stem cells. While another research direction for “bedside to bench” will be to address the problems encountered during clinical treatment of cancer patients, such as drug resistance, cancer metastasis, cancer recurrence, serious complications resulted from treatment etc, in order to develop personalized medicine and executed by the ‘Cancer Research Center of Excellence’ of both CMU&H and NTUCM. Specific Aims: 1. To discover the novel mechanisms of tumorigenesis, metastasis and drug resistance of cancers with common interests, specifically focus on the endemic cancers with ethnic differences between East and West 2. To identify druggable pathways and targets 3. To develop biomarkers and effective therapies to improve treatment outcome of cancer patients
