    Title: 台灣產山螞蝗屬及其近源植物抗氧化活性及其神經細胞保護作用機轉之探討;The antioxidant activity and neuroprotective effects of Desmodium genus and related plants in Taiwan

    Abstract: 巴金森氏症為老年常見神經退化性疾病之一,其致病因素與體內過多自由基所產生的氧化壓力有關。近年來對於巴金森氏症之研究指出,MAO-B 抑制劑及抗氧化劑對巴金森氏症之神經損傷具有保護作用。本實驗選用 4 種山螞蝗屬植物及其同為豆科蝶型花亞科之3 種近源植物為實驗材料,進行抗氧化成分含量、抗氧化活性及MAO 抑制活性之評估;並進一步探討其對 6-OHDA 誘發 SH-SY5Y 神經細胞株細胞損傷之保護作用機制。
    首先測定七種植物甲醇抽取物抗氧化活性成分總多酚類、黃酮類及苯基丙酸之含量,結果發現以排錢樹、波葉山螞蝗及鐘萼豆含量最多,而大葉山螞蝗含量最低。其次,在抗氧化活性上,亦以排錢樹、波葉山螞蝗及鐘萼豆具有較佳之清除 DPPH 自由基能力、總抗氧化活性及還原能力;而大葉山螞蝗及三點金草則為七種植物中活性最低者。至於,在超氧陰離子清除能力上,仍以排錢樹、波葉山螞蝗及鐘萼豆具較佳活性;但在過氧化氫及氫氧自由基之清除能力上,則分別以鐘萼豆及葫蘆茶具較佳活性。另在抑制單胺氧化酶之活性上,則分別以鐘萼豆及波葉山螞蝗對 MAO-A 及 MAO-B 具較佳之抑制活性,且以波葉山螞蝗對 MAO-B 較具選擇性。
    接著,進一步探討具較高抗氧化活性之排錢樹、波葉山螞蝗、鐘萼豆及葫蘆茶對 6-OHDA 誘發 SH-SY5Y 神經細胞氧化損傷之保護機制。實驗結果發現,排錢樹、鐘萼豆及葫蘆茶均可提高 6-OHDA 誘導 SH-SY5Y 神經細胞損傷後之存活率,但波葉山螞蝗則無保護作用;其作用機轉可能與其抗氧化活性及能有效降低細胞內氧化壓力以降低細胞損傷有關,而活性成分則可能為總酚類成分。

    Parkinson’s disease, one of widespread neurodegenerative disorders, is related to intercellular oxidative stress caused by overproduction of free radical. Therefore, recent researches indicated that MAO-B inhibitor and antioxidants may prevent Parkinson’s disease. This study was aimed to investigate the antioxidant components, anti-oxidative activity and MAO inhibiting effects of four specise of Desmodium plants and three related plants by microtiter spectrophotometric and fluorescent methods.
    First, the experimental results demonstrated that Phyllodium pulchellum, Desmodium sequax and Codariocalyx motorius have enriched antioxidant components including total phenol, flavonoid, and total phenylpropanoid glycoside. These above three plants also have the better antioxidative capacity than the others. There are a very closely correlation among total phenols content, Trolox equivalent antioxidant capacity and reducing power of the selected seven plants.
    Second, Phyllodium pulchellum, Desmodium sequax and Codariocalyx motorius have the better superoxide anion radical scavenging activity. The Codariocalyx motorius and Tadehagi triquetrum have better hydrogen peroxide and hydroxyl radical scavenging activity than the others. On the other hand, Codariocalyx motorius and Desmodium sequax have better MAO-A and MAO-B inhibiting effects, and Desmodium sequax has better selectivity in inhibiting MAO-bactivity.
    Furthermore, Phyllodium pulchellum, Desmodium sequax, Codariocalyx motorius, Tadehagi triquetrum, possessing the better antioxidant potency were selected to compare the neuroprotective effects on 6-OHDA induced oxidative damage in SH-SY5Y neuroblastoma cell.
    The present result demonstrated that Phyllodium pulchellum, Codariocalyx motorius, and Tadehagi triquetrum can elevate cell viability, but Desmodium sequax can not. Phyllodium pulchellum, Codariocalyx motorius, and Tadehagi triquetrum protected the 6-OHDA-induced oxidative damage in SH-SY5Y neuroblastoma cell via decreasing intracellular oxidative stress and have better antioxidative.
