    Title: 熟地黃萃取物治療阿茲海默氏症的電腦輔助藥物設計之研究

    Abstract: 阿茲海默氏症,一般又稱為癡呆症,是一種退化性的神經疾病,主要的特徵有顯著記憶力減退並且會逐步惡化、不正常的語言能力、性格也會改變以及認知功能惡化、無法照顧自己。現在已知,患者的大腦會記憶力減退以及他們的乙醯膽鹼的分泌作用和神經細胞的減少有密切相關,造成患者腦中乙醯膽鹼的量和大約90 %的正常人比較起來是有落差的。
      在這個實驗中,中草藥熟地黃對阿茲海默氏症有效的成分,和目前最有效的治療阿茲海默氏症的乙醯膽鹼合成酵素抑制劑做比較。因此,我們想要研究藥用熟地黃當成主要的藥物,熟地黃藥物的結構和阿茲海默氏症在乙醯膽鹼的酵素結構在實驗中熟地黃抑制乙醯膽鹼的效用,實驗用程式Discovery Studio 軟體將主要用乙醯膽鹼酵素和接合的熟地黃分子之間結構藉由對接的程式模擬並分析計算此藥物的活性。實驗結果熟地黃梓醇藥物活性的評估獲得不錯的分子接合分數,所以得到最初目的的實驗結果,未來實驗將會更進一步探究熟地黃其他成分的結構是否也能有乙醯膽鹼酵素抑制作用,在不久的將來盡快研究這些成分來確認。
    Alzheimer's disease, which is called dementia in general is one kind of brain degenerative nerve disease, mainly characterized by significant and progressive deterioration of memory loss, abnormal language skills, personality changes and deterioration of cognitive function cannot take care their own. As previous study reveals that the brain of patients with memory loss and their secretion of acetylcholine are related to the decrease of nerve cells, making levels of the brain acetylcholine in patients compared with 90% normal difference.
    In this study, the ingredients of Chinese herbs Shu di Huang for the efficacy of Alzheimer's disease, and compared with the chemical acetylcholine synthesis inhibitor. Therefore, we want to study medicine Shu di Huang as the main drug, the drug Shu di Huang structure and Alzheimer's disease in the enzyme structure of acetylcholine on the experiment of Shu di Huang inhibitory effect of acetylcholine, Discovery Studio? experimental program will mainly use acetylcholine Shu di Huang molecular structure of the enzyme and the binding between the simulation and analysis using Docking program to evaluate its drug activity. The experimental results obtained show that component of the preliminary test results Shu di Huang catalpol the assessment of drug activity to obtain a good docking score, future experiments will further explore whether the structure of other components of Shu di Huang is also able to acetylcholine enzyme inhibition, to be confirmed as soon as these components can be studied in the near future.
