    題名: 以鮪魚蒸煮液製備抗癌活性胜?及其生物利用性之評估

    摘要: 生物活性胜? (bioactive peptides) 具有促進小腸細胞增生、調節免疫、抗菌、抗 血栓形成、降血壓、抗癌等保健功效。魚肉蛋白水解物 (fish protein hydrolysate; FPH) 中 之生物活性胜?已被證實具有良好之抗氧化能力,包含清除自由基能力、還原力、螯 合金屬離子活性等,另對於降血壓亦有效果。過去對於生物活性胜?抑制腫瘤或癌症 的研究發現,大豆蛋白水解物有抗腫瘤及抗癌之效果,亦具有抑制癌細胞轉移及侵入 之功效;但魚肉蛋白水解物中具有抗癌功效之研究目前已知的只有由鯷魚醬油之丁醇 抽出物對人類淋巴癌細胞有抑制增生 (antiproliferation) 效果;另外多種魚種之魚肉蛋 白水解物對人類乳癌細胞亦有抑制增生的效果,顯示水解物中之胜?應具有抗癌功 效,但以上研究並未進行後序胜?分離及純化,另胺基酸序列亦無鑑定。因此魚肉蛋 白水解物之抗癌功效尚缺乏足夠之研究加以證明,且其胜?之純化及其胺基酸組成及 序列亦需鑑定。 本研究室經預備實驗發現,鮪魚蒸煮液蛋白質水解液具有抑制乳癌細胞增生的功 效,因此本研究擬利用商業蛋白?,對鮪魚蒸煮進行水解,製備並鑑定具抗癌活性之 生物活性胜?之胺基酸序列,另以模擬消化及吸收系統,評估其生物利用性,期望能 開發成保健食品並減少環境污染、提升加工副產物之經濟價值。

    The identification of growth factors, immunomodulating, antimicrobial, antithrombotic, hypotensive and anticancer peptides from milk, wheat, soybean and egg is a scientific evidence that enzymatic hydrolysis of food proteins is an efficient way to recover potent bioactive peptides. Also, research on fish protein hydrolysates (FPH) demonstrated they constitute a source of antioxidants which possessed 2,2-diphenyl-1-pycrylhydrazyl (DPPH) and hydroxyl radical-scavenging capacity, reducing power, chelating metal ion and antihypertensive activities. Researchers demonstrated that soy protein hydrolysates showed anticancer, antitumor activities and inhibition of metastasis and invasion of cancer cell lines. To date, only two studies were done for the anticancer activities of FPH. The first one is that the peptides from anchovy sauce induced apoptosis and antiproliferation in a human lymphoma cell (U937). The second one is that FPH induced the antiproliferative activity in human breast cancer cell lines. However, the later study did not further isolate and purify the bioactive peptides, and both studies did not determine the amino acid sequence of the peptides. Therefore, the knowledge of the anticancer activity of FPH is limited, and the detail information (such as the amino acid sequence) needs further researches to be determined. According to the preliminary data, hydrolysates from tuna cooking juice possessed antiproliferation activity of MCF-7 cell line. Therefore, the objective of this study is to utilize different types of commercial proteases to hydrolyze the proteins of tuna cooking juice, and to produce the anticancer peptides and determine their peptide sequence. Then, the bioavailability of the anticancer peptides is determined using simulated gastrointestinal digestion and Caco-2 cell permeability. We try to develop the functional food with anticancer activity to elevate the economic values of by-products from a tuna processing factory and reduce environment pollution.
