摘要: | 探討植物肉桂中生物活性成分α-水芹烯誘導小鼠血癌和人?血癌細胞株(WEHI-3 與 U937)產生細胞凋亡與正常和血癌小鼠中免疫調控之?體與活體之研究 α-水芹烯(α-phellandrene),為辛香?及香草植物中常?之活性成份,如:中藥中的肉桂、生薑、 ?薄荷、艾?、大蒜、細辛、松?……等,且許多文獻指出其有許多生物活性的效果。但目前α-水芹 烯對血癌則無相關之?體與活體之抗癌活性研究。因此,本計畫之研究目的主要在探討α-水芹烯對小 鼠血癌細胞株WEHI-3 及人?血癌細胞株U937 進??體(in vitro)及活體(in vivo)抗血癌評估及免疫調 節的研究。擬分為三主題並預期三?內完成。第一?計劃目的是:(1) 探討α-水芹烯對小鼠血癌細胞 株(WEHI-3)之細胞週期停滯及細胞凋亡的影響。(2) 探討α-水芹烯對WEHI-3 細胞對於細胞週期停滯 和細胞凋亡相關蛋白質與基因層次的影響。(3) 建?BALB/c 小鼠血癌模式,檢測α-水芹烯對於抗 WEHI-3 小鼠血癌之抗血癌評估。(4) 檢測α-水芹烯對於抗血癌小鼠之免疫調節之評估。第二?計劃 目的是:(1) 檢測α-水芹烯對人?血癌細胞(U937)之細胞週期停滯及細胞凋亡相關?徑的影響。(2) 檢 測α-水芹烯對U937 細胞對於細胞凋亡相關蛋白質與基因層次(內在?徑、外在?徑與內質網壓?)的 影響。(3) 檢測α-水芹烯對於人?血癌U937 ?種移植於BALB/c ?小鼠腫瘤之影響評估。(4) 檢測α- 水芹烯對於?種移植腫瘤之凋亡相關蛋白質與基因層次評估。第三?計劃目的是:?用正常小鼠、? 小鼠(T 細胞缺陷)與?化小鼠進?α-水芹烯對小鼠免疫調節之影響,評估項目如:(1) 小鼠外觀與重? 評估。(2) 小鼠週邊單核球細胞評估(細胞族群評估、血清中細胞激素與吞噬細胞吞噬活性)。(3) 小鼠 脾臟細胞評估(T 與B 細胞增殖、自然?手細胞毒?活性和細胞激素檢測)。(4) 小鼠血清中,生化檢測 評估安全性。此??藉由這些檢測進??解α-水芹烯在提升正常小鼠、免疫功能缺失小鼠與?齡化小 鼠免疫功能之探討。
Investigations of α-phellandrene, a bioactive compound from Cinnamon, in induction of apoptotic cell death in murine and humam leukemia cell lines (WEHI-3 and U937) and immune modulation in normal and leukemia mice in vitro and in vivo. Alpha-phellandrene is a common and active compound from spicy and aromatic plants including traditional Chinese medicine cinnamon, ginger, green mint leaves, garlic, Asarum, pine needles and so on. It is reported that α-phellandrene exhibited many biological activities; however, there is no available information addressing α-phellandrene-affected biological and anticancer activity on leukemia in vitro and in vivo. Therefore, the purpose of this project was to investigate the α-phellandrene on the murine leukemia cell line (WEHI-3) and human leukemia cell line (U937) in vitro, anti-leukemia activity and immunemodulations in vivo. This proposal can be divided into 3 subjects and should be finished within three years. Subject I in the first year is to investigate 1) the effects of α-phellandrene-induced cell cycle arrest and apoptosis in murine leukemia WEHI-3 cells in vitro; 2) the effects in cell cycle arrest and apoptosis-associated protein and gene expression levels in WEHI-3 cells; 3) estabolishment of leukemia mice and investigations of α-phellandrene-affected leukemia in vivo; 4) influences of immune response in leukemia mice in vivo. Subject II in the second year is to investigate 1) the effects of signaling pathways in α-phellandrene-induced cell cycle arrest and apoptosis in human leukemia U937 cells in vitro; 2) the effects in apoptosis-associated protein and gene expression levels (extrinsic pathway, intrinsic pathway and ER stress) in U937 cells; 3) investigations of α-phellandrene-affected murine xenograft leukemia transplantation tumor in vivo; 4) influences of apoptosis-associated protein and gene expression levels from transplantation tumor after α-phellandrene exposure in vivo. Subject III in the third year is to investigate the effects of immune responses in α-phellandrene-treated normal mice, nude mice (T-cell deficiency) and aging mice in vivo. Assessment projects such as: 1) appearance and weight of mice; 2) assessment of peripheral mononuclear blood cells including cell population, cytokines serum and phagocytic activity; 3) determinations of splnocytes from mice, such as T and B-cell proliferations, natural killer cell cytotoxic activity and cytokine; 4) biochemical examinations in serum is to assess safety. Based on these determinations in this year, it is known that α-phellandrene promoted the immune responses in the normal mice, immunodeficiency mice and aging mice in vivo. |
