    摘要: Histidine為人體必需胺基酸之一,存在於腦、骨骼肌、肝臟中。已有研究顯示其具有抗氧化、抗發炎等保護作用。本實驗室之前研究顯示,飲水中0.1% histidine或canosine的添加可降低餵食5%奶油飲食的小鼠其發炎及氧化傷害。由於添加histidine與carnosine兩者有相似的效果,而carnosine價位較高,因此本研究將探討於給予不同濃度的histidine飲水,對於餵食15%奶油飲食的小鼠之影響。本實驗使用4週齡大的C57BL/6小鼠,分成 (1)正常組:餵食一般飲食 (2)控制組:為添加15%奶油飲食及一般飲水 (3)0.1% histidine組:15%奶油飲食與0.1%的histidine飲水 (4)0.2% histidine組:15%奶油飲食與0.2%的histidine飲水。餵食8週後犧牲,並測定體重、血糖、副睪脂重、胰島素、體脂質、尿酸、xanthine oxidase與脂質代謝相關荷爾蒙。實驗結果發現,餵食15%奶油飲食可以顯著增加體重、副睪脂重、胰島素、心臟、肝臟和腎臟的尿酸、血清和肝臟三酸甘油酯及總膽固醇、肝臟LDL,賀爾蒙方面有血清leptin濃度提高,血清ghrelin與adiponectin濃度下降。而給予histidine的飲水後,顯著降低小鼠血清insulin、HOMA-IR值與尿酸濃度;也顯著降低血清膽固醇、肝臟三酸甘油酯、LDL及leptin濃度;也提高血清ghrelin及adiponectin濃度。實驗結果推測histidine的補充,可能可以減緩因攝取大量飽和脂肪所造成的高血脂及胰島素阻抗,進而降低代謝症候群的發生。
    Histidine is an essential amino acid and present in brain, skeletal muscle and liver. Several studies have reported that it could provide anti-oxidative and anti-inflammatory protection. Our previous study also found that histidine or carnosine supplement markedly reduced oxidative and inflammatory stress in mice consumed 5% high saturated fat diet (HFD). Furthermore, we observed that histidine and carnosine exhibited similar effects. The major purpose of this present study was to investigate the effects of 0.1% and 0.2% histidine in mice consumed 15% HFD prepared from butter. Mice were divided into 4 groups: normal diet with water, 15% HFD with water, 15% HFD with 0.1% histidine and 15% HFD with 0.2% histidine for 8 weeks. Results showed that mice consumed HFD had higher body weight, epididymal fat weight, insulin, uric acid, triglyceride (TG), total cholesterol (TC), low density lipoprotein and leptin levels; and lower ghrelin and adiponectin levels than mice with normal diet. However, the intake of histidine significantly diminished level of insulin, HOMA-IR, uric acid, liver TG, LDL, serum TC. Supplementation of histidine also significantly reduced leptin, and increased ghrelin and adiponectin. These findings suggested that histidine was a potent agent to alleviate high fat diet induced hyperlipidemia and insulin resistance.
