    題名: 落新婦等8種藥用植物抗氧化活性暨落新婦之藥理活性評估
    Studies on the Antioxidant Activities of Eight Medicinal Plants and Evaluations of Pharmacological Activities of Astilbin longicarpa

    摘要: 本研究,針對八種台灣具鎮痛抗發炎活性的藥用植物,包括:落新婦(虎兒草科)、長果藤(苦苣苔科)、角桐草(苦苣苔科)、睫穗蓼(蓼科)、栗蕨 (碗蕨科)、黑果馬皎兒(瓜科)、山澤蘭(菊科)、千里光 (菊科),進行甲醇萃取,再以其萃取物進行其抗氧化活性比較,分析方法包含:1. DPPH自由基清除效應、2. Trolox當量的總抗氧化能力測定、3. 還原力測定、4. Ferric reducing卅antioxidant powder(FRAP),亦測定其總多酚類、類黃酮類與黃酮醇類成分含量,分析其實驗結果並加以探討,結果發現虎耳草科落新婦之抗氧化活性尤佳,其總多酚、黃酮類成分含量亦高,進而對落新婦進行更進一步的研究 。
    將落新婦甲醇萃取物依次以Hexane、乙酸乙酯、正丁醇進行液相-液相萃取,得到Hexane層、乙酸乙酯層、正丁醇層和水層等產物,再以其各層萃取物進行抗氧化活性比較,並以HPLC評估落新婦指標成分Bergenin含量,又因近年來文獻指出落新婦具有抗腫瘤活性,便以Hexane層、乙酸乙酯層、正丁醇層和水層分別針對人類非小細胞肺癌細胞株 A549進行細胞毒殺性試驗。
    研究結果顯示落新婦塊根乙酸乙酯層有最佳之抗氧化能力,由HPLC分析的結果,正丁醇萃取層之Bergenin含量最高,此外,在細胞毒殺性試驗中,落新婦塊根乙酸乙酯層與落新婦地上部位乙酸乙酯層對類非小細胞肺癌細胞株A549具有良好的致死效果。因此台灣特有種落新婦活性成份的分離、純化及作用機轉之探討,值得進一步研究。
    The aim of this study was to evaluate antioxidant activities of eight medicinal plants including Astilbe longicarpa (Hayata) Hayata, Aeschynanthus acuminatus Wall. ex A. DC, Hemiboea bicornuta (Hayata) Ohwi, Polygonum longisetum De Bruyn, Pteris incisa Thunb., Zehneria mucronata (Blume) Miq, Eupatorium chinense L. var. tozanense (Hayata) Kitam., and Senecio scandens Ham. ex D. Don. Four scavenging effects were determined including: 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging activity, trolox equivalent antioxidant capacity (TEAC), reducing power, and ferric reducing/antioxidant power (FRAP). We also analyzed their phenolic compounds, flavonoids, and flavonol contents.

    The extracts of all eight plants were found to present certain radical scavenging and antioxidant activities. In particular, Astilbe longicarpa (Hayata) Hayata exhibited best antioxidant activity and contained higher quantity of polyphenols and flavonoids. Therefore, Astilbe longicarpa (Hayata) Hayata was further investigated.

    We evaluated the antioxidant activity of MeOH extract and sequential extracts of n-hexane, ethyl acetate, and n-BuOH from the dried aerial parts and rhizomes of Astilbe longicarpa. Four fractions were compared for their antioxidant capabilities and HPLC methods were also applied to analyze the contest of the major active component - bergenin. Furthermore, the MTT assay was performed to measure the inhibition on A549 human lung cancer cell for it’s anticancer effects.

    Results showed that crude ethyl acetate extract of Astilbe longicarpa (Hayata) Hayata exhibited higher free radical scavenging activity and performed stronger inhibitive effect on A549 human lung cancer cell. Therefore, the isolation, purification, and the mechanism of active components in Astilbe longicarpa (Hayata) Hayata need to be further investigated.
