    Title: 豬苓多糖對化療藥物誘發之遺傳損傷的防護作用

    Abstract: 探討豬苓多糖對化療藥物Cyclophosphamide(CP)所導致遺傳損傷的防護作用 方法:在體外實驗使用人類淋巴胚母細胞株 (human lymphoblastoid cell line;TK6),投予新鮮泡製豬苓多糖溶液300 g/ml, 30分鐘後再分別暴露於80 g/ml或120 g/ml劑量的CP,經30分鐘去除豬苓多糖及CP,再培養48小時後,於螢光顯微鏡下計數染色體損傷微核的生成率;體內實驗以6~8週ICR公鼠,腹腔注射豬苓多糖4.0 mg/kg、 12.5 mg/Kg 和50 mg/Kg的劑量,於30分鐘後給予50mg/Kg劑量的CP,並於CP給藥後48小時採鼠尾血計數微核。 結果:實驗證明,不管體外、體內微核試驗,CP可使染色體損傷而導致微核增加,經由豬苓多糖事先投予,再暴露於CP後測定微核量,結果顯示豬苓多糖於體外、體內試驗統計上均能有意義的降低CP誘發的微核數,使我們更確定豬苓多糖對於CP所誘發的遺傳損傷具有保護能力。 結論:體外、體內微核試驗結果顯示,豬苓多糖其本身是個不具細胞毒性且有很好化學防護作用的中藥材成分。
