    Title: 探討幽門螺旋菌細胞毒素相關基因A與脂質筏之相關

    Abstract: 幽門螺旋菌(Helicobacter pylori)被發現是一種重要持續性感染的致病菌已經有二十年了。在大多數感染的個體中,幽門螺旋菌僅導致慢性胃發炎而且通常是無症狀的。但如果是長期持續性的感染,有部分個體可能會引起胃潰瘍與十二指腸潰瘍,甚至些患者還會導致胃癌或淋巴瘤。我們先前的研究著重於細胞脂質筏(lipid rafts)在幽門螺旋菌侵入上皮細胞所扮演的角色,我們確認幽門螺旋菌侵入上皮細胞與其脂質筏有關,但是與細菌的黏附能力無關。我們亦觀察到幽門螺旋菌侵入細胞之處有膽固醇的聚集。更進一步證明細菌的致病因數VacA(vacuolating cytotoxin gene A)、CagA(cytotoxin-associated gene-endoded protein)與BabA2(blood-group antigen adhesion)皆會與脂質筏作用,而且可能進一步提供脂質筏叢聚的功能。本實驗亦將深入探討CagA蛋白質與脂質筏交互作用的關係,我們將cagA基因轉殖進入細胞藉以表現CagA蛋白質,並利用共軛焦顯微鏡(confocal microscope)來觀察CagA與細胞分子之交互作用,期能更清楚瞭解幽門螺旋菌導致宿主細胞之致病機轉,而且將來在臨床上,更可以促進發展新的策略來防止細菌的持續性感染。
