    Title: 分析人類腸道分離菌之-Glucosidase活性與其最適作用條件

    Abstract: 大部份的中草藥以口服的方式給藥,而其中部份成份經胃腸道菌叢的作用,轉變為具療效或較易為人體吸收之有效成分,胃腸道菌叢所表現之-glucosidase對轉換中草藥中有效成分,使之易於吸收扮演重要角色。我們利用Geniposide與-Glucosidase反應,使Geniposide產生水解作用而產生Genipin,再利用胺基酸與Genipin作用。我們利用此一產生色素的機制來篩選人類腸道分離菌的-Glucosidase活性與其最適作用條件。在這篇研究裡,我們先將5株人類腸道分離菌株,以MRS 培養基或培養液做初步篩選,經過適當的稀釋倍數後加入Geniposide及Glycine與其分別反應12、18、24小時的時間,並經過調整pH值到5.5持續培養2小時後,在經過80℃高溫培養1小時後,測得其所產生的色素的OD值,以推算何者產生較佳的-Glucosidase活性。最後得知Lactobacillus ssp. JB3於培養18小時所產生之後,表現最佳之-Glucosidase活性。以LB及BHI培養基培養Lactobacillus spp. JB-3,可得較MRS培養高2.4倍及1.3倍之β-Glucosidase活性。
