    題名: 冬瓜子調節血糖之保健功效研究

    摘要: 糖尿病的發生在全世界已成為主要嚴重威脅人類健康的慢性疾病之一。在台灣,糖尿病已成為國人十大死因之一。糖尿病患者常需要長期藥物的使用(例如胰島素),來達到理想血糖控制,而避免糖尿病所引起之相關併發症並改善病人生活品質。然而,目前已有報導指出部分抗糖尿病製劑並不能夠有效的去改善糖尿病患者高血糖的情形;或是產生與治療無關且有害的副作用。因此,發展更有效且安全的降血糖保健食品來幫助糖尿病患者是非常重要的課題。冬瓜(Benincasa hispida (Thunb). Cogn)是屬於葫蘆科一年生蔓性植物。在中國、印度和其他的一些亞熱帶國家是一種普遍使用的蔬菜。在中國和韓國中醫學認為冬瓜味甘而性寒,有利尿消腫、清熱解毒、清胃降火及消炎之功效,因此冬瓜成熟之後的果肉(fruit)和種子(seed)的萃取物,在中醫主要是用來治療動脈硬化、冠心病,高血壓、水腫腹脹、糖尿病和消渴症等疾病。然而,冬瓜子(Semen Benincasae)萃取物對於抗高血糖和治療糖尿病的效應並不清楚。對此,希望可以透過本研究計畫,來進一步探討冬瓜子萃取物[Semen Benincasae extract (S.B.E.)]對於調控血糖和治療糖尿病的保健功效與確切的作用機制。我們的主要研究目標為:(1)利用正常鼷鼠飢餓模式與高血糖鼷鼠模式(高脂肪飼料誘導的第二型糖尿病模式),研究冬瓜子萃取物在動物體內的效應,包括:血糖的調控、血漿中胰島素的濃度、以及其相關的作用,如:口服葡萄糖耐受性試驗和胰島素耐受性試驗。(2)利用胰臟beta細胞衍生的RIN-m5F細胞株探討冬瓜子萃取物調節血糖功效的可能機制。 冬瓜子是經常被使用的農產品之ㄧ,在日常生活中很容易就可以取得,我們希望經由以上的實驗,能夠確認冬瓜子萃取物對於糖尿病鼷鼠改善高血糖症狀和調控血漿胰島素分泌的影響,並且了解冬瓜子對於調節血糖的保健功效機制為何。希望透過本研究的科學化證據,讓冬瓜的農產價值提升,並使冬瓜子能夠成為具有功效且安全的糖尿病保健食品。

    Diabetes mellitus (DM) is growing in prevalence worldwide and is becoming a serious threat to the main kind of health in the worldwide. In Taiwan, diabetes mellitus is one of the top ten deaths. Diabetic patients need pharmacological help to achieve optimal glycemic control to preventing diabetic complications and improving quality of life. However, it has been reported that some of anti-diabetic drugs (sulphonylureas and biguanides) were not ability to significantly alter the rate of progression of hyperglycemia in patients with type-2 diabetes. Therefore, there is urgently needed for developing more effective, advantageous, and safe anti-hyperglycemic agents to protect our health. Benincasa hispida (Thunb). Cogn (Syn. Benincasa cerifera), which belongs to a family of Cucurbitaceae, is a kind of vegetable used widely in Taiwan, China India and other semi-tropical countries. In Chinese and Korea, Benincasa hispida mature fruit or seed extract (called: Semen Benincasae or Dong Gua Zi) was used mainly diabetes and diuresis diseases. However, the effects of S. Benincasae extract on anti-hyperglycemia or anti-diabetic effect are still unclear. In this research project, it will carry out to evaluate and understand the precise action mechanism of anti- anti-hyperglycemia or anti-diabetic effect of Semen Benincasae. We will propose specific aims: (1).Using animal model [fasted normal mouse model and high fat diet-induced diabetic mouse model (type-2 diabetic model)], to study the in vivo effects of S. Benincasae extract on blood glucose regulation, plasma insulin levels, and relative effects (oral glucose tolerance test (OGTT) and insulin tolerance test (ITT)). (2).to investigate the blood glucose regulatory effects of S. Benincasae extract on pancreatic islet beta-cell line RIN-m5F and its possible involved molecular mechanisms. The plant of Benincasa hispida (Thunb). Cogn (Syn. Benincasa cerifera) is a kind of vegetable which used widely in Taiwan, and easily to obtain in our daily life. In this project, we hope to identify effects of improving hyperglycemia, regulating of plasma insulin secretion and protective efficacy of health in hyperglycemia mice. Identifying and achieving these scientific evidences can more elevate agricultural and economic value of Benincasa hispida, and worthy of developing an advantage and safe health food in diabetes.
