    Abstract: 本計畫將依過去的經驗、利用 glutaldehyde方法將streptomycin直接接上蛋白質以供免疫動物產生抗體, 並依同棣方法將streptomycin接上酵素以供檢定所產生的抗體的力價及其用於ELISA檢測方法的可行性.Erythromycin的衍生物則將依產生oxime的方法將其變成衍生物、再依isochloroformat的方法將其結合上蛋白或酵素HRP以供檢定抗體的力價及其他建立ELISA的條件.建立Streptomycin及ELISA的條件包括標準曲線、準確度、敏感度、recovery rate檢體的檢驗及利用毛細管電泳及HPLC確認ELISA的可靠性.

    This research project attempts to create Streptomycin derivative by glutaldehde method and then conjugate this derivative to a protein for immunization of animal to produce antibody against streptomycin.The conjugation of streptomycin derivative to enzyme, HRP, will also be carried out for the titration of antibody and establishment of ELISA.Erythromycin will be converted to derivative by oxime method by utilizing two keto groups in the erythromycin.The derivative will then be conjugated to protein for immunization and to enzyme, HRP, for titration of antibody and formulation of ELISA kits.The validation of ELISA for both streptomycin and erythromycin will be carried according to our past experience in establishing standard curve, sensitivity determination, cross-reactivity test, recovery rate and utilizing HPLC and/or capillary electrophoresis to verify and validate the ELISA.
