    摘要: 本研究首先探討倒地蜈蚣甲醇粗抽物及其成分betulin於醋酸扭體法及福馬林疼痛試驗法之鎮痛作用,並以λ-角叉菜膠誘導大鼠足蹠腫脹,來探討其抗發炎作用,最後並測定大鼠肝組織及紅血球之超氧歧化酶、麩胱甘肽還原酶、及麩胱甘肽過氧化酶活性、發炎足蹠組織之脂質過氧化指標丙二醛、與一氧化氮之含量變化,以探討倒地蜈蚣是否藉由提升抗氧化酵素之活性而達到抗發炎作用。 鎮痛抗發炎實驗測定,倒地蜈蚣甲醇粗抽物 (1.0, 2.0 g/kg)及其成分betulin (30, 90 mg/kg)均可減少由醋酸所引起疼痛之扭體次數。而在福馬林引起之疼痛反應,倒地蜈蚣甲醇粗抽物(2.0 g/kg)及其成分betulin(10, 30與90 mg/kg)均可明顯減少小鼠福馬林引起之early phase及late phase之舔足反應時間。倒地蜈蚣甲醇粗抽物 (0.5, 1.0與2.0 g/kg)及其成分betulin (30, 90 mg/kg),可明顯抑制λ- carrageenan誘導的急性足蹠腫脹。 肝組織抗氧化酵素活性測定,倒地蜈蚣甲醇粗抽物及其成分betulin對肝臟中之SOD、GR 及GPx活性均具增強作用。紅血球抗氧化之相關測定,倒地蜈蚣甲醇粗抽物倒地蜈蚣甲醇粗抽物及其成分betulin對紅血球中GR及GPx的活性均具增強作用。而倒地蜈蚣甲醇粗抽物(0.5, 1.0和2.0 g/kg)及其成分betulin(30, 90 mg/kg),亦均會減少大鼠發炎後足蹠MDA的含量,以降低λ- carrageenan所引起的發炎反應。Betulin(30, 90 mg/kg)亦會減少大鼠誘導發炎後足蹠NO的含量。 綜合以上結果顯示,鎮痛實驗中:倒地蜈蚣甲醇粗抽物及其成分betulin對於醋酸所引起的扭體反應均具有抑制作用。在福馬林所誘發的疼痛反應,倒地蜈蚣甲醇粗抽物及其成分betulin在中樞及周邊,均產生明顯的抑制作用。而於抗發炎實驗中,倒地蜈蚣甲醇粗抽物及其成分betulin均可減少λ-carrageenan誘發之足蹠腫脹體積,顯示其具有抗發炎作用。其作用機轉可能與提升肝臟及紅血球中抗氧化酵素之活性或減少大鼠足蹠腫脹發炎後之MDA的含量有關。而成分betulin更能抑制NO的含量,而達抗發炎作用。; In the present study, the analgesic effect of Torenia concolor Lindley var. formosana T. Yamazaki (TC) and betulin, an active composition of TC, were investigated by acetic acid-induced writhing response, formalin test, and the anti-inflammatory effect against λ-carrageenan-induced paw edema. In order to understand the mechanism of anti-inflammation of TC, superoxide dismutase (SOD), glutathoine reductase (GR) and glutathione peroxidase (GPx) levels in blood and liver, and malondialdehyde (MDA) and nitric oxide (NO) in the rats paw were measured. The results shown that methanol extract of TC (TCMeOH, 1.0, 2.0 g/kg) and betulin (30, 90 mg/kg ), significantly inhibited the acetic acid-induced writhing response. TC MeOH (2.0 g/kg) and betulin (10 , 30 and 90 mg/kg) significantly inhibited formalin-induced licking time during the early phase and late phase. TC MeOH (0.5, 1.0 and 2.0 g/kg) and betulin (30, 90mg/kg) also significantly decreased the λ-carrageenan-induced paw edema. In addition, TCMeOH and betulin treatment also caused significant increase in SOD, GR and GPx levels in liver, and GR and GPx levels in erythrocytes, and decrease in the MDA level in paw. Furthermore, betulin also caused significant reduction of NO level in paw. These findings support the view that TCMeOH and betulin exert analgesic and anti-inflammatory effects. Effects of TC MeOH and betulin on carrageenan-induced paw oedema may be related to its anti-oxidant effect in the liver and erythrocytes and decreasing the MDA production in paw. Betulin also inhibited NO production.
