天然產物應用於癌症化療已有三十多年歷史。本論文所用之3-Hydroxycinnamic acid是咖啡酸的衍生物。研究主題在評估3-Hydroxycinnamic acid 對人類子宮頸癌細胞株(HeLa cells)引發的反應,包括對細胞生長的抑制作用,細胞群的週期變化,以及誘導細胞凋亡並探討其機制。結果顯示3-Hydroxycinnamic acid對人類子宮頸癌細胞株(HeLa cells)有劑量性(dose-dependent)之細胞生長抑制作用,利用流式細胞儀和DNA膠體電泳分析,也顯示對該細胞株會引起細胞凋亡作用。進一步利用西方墨點法檢測出細胞週期調控蛋白cyclin B1,cyclin A,P53表達量增加。因此我們認為P53的明顯增加這極可能是該藥物引發HeLa cell凋亡之重要機制之一。; Natural products have been the mainstay of cancer chemotherapy for the past 30 years. 3-Hydroxycinnamic acid is one derivatives of Caffeic Acid (CAF). In this study, we report that treatment with 0.1mM ,0.5mM ,1mM , 2mM , 5mM , 10mM 3-Hydroxycinnamic acid induced dose-dependent cytotoxicity and apoptosis in human cervical cancer cell line (HeLa cells). Cells showed the typical DNA laddering pattern in DNA gel electrophoresis and other characteristics of apoptosis with flow cytometric analysis. Further ,western blotting analysis showed that the level of cyclin B1 ,cyclin A and P53 were increased after treatment with the 3-Hydroxycinnamic acid for 24hr. We suspect that P53 may play an important role in 3-Hydroxycinnamic acid induced apoptosis in HeLa cells.
