    题名: 中草藥對抑制黑色素生合成與抗氧化活性之研究;Studies on Inhibition of Melanogenesis and Antioxidant Capacity of Chinese Herbs

    摘要: 中文摘要 本研究分別以酪胺酸酶活性試驗,多巴色素自動氧化生成黑色素量試驗和擬超氧化物歧化酶活性試驗,評估15種中草藥(積雪草、毛果延命草、黃花敗醬草、山豆根、台灣木通、台灣蛇莓、小萹蓄、虎耳草、銳葉楊梅、紫蘇葉、絲瓜葉、毛梗豨薟草、檳榔、胡椒、荖葉)乙醇萃取物對抑制黑色素生合成與抗氧化之活性。體外試驗結果顯示,在對酪胺酸酶活性試驗中,低濃度(50µg/ml)時,以虎耳草對酪胺酸酶具較明顯的抑制作用(42.9±0.5%);中濃度(200µg/ml)時,以積雪草對酪胺酸酶較具明顯的抑制作用(44.9±0.4%);高濃度(500µg/ml)時,以胡椒對酪胺酸酶具較明顯的抑制活性(41.0±0.5%)。然而在不同濃度測試中,毛果延命草低濃度(50µg/ml)時則對酪胺酸酶具抑制活性,中濃度(200µg/ml)時則促進其活性,高濃度(500µg/ml)時對酪胺酸酶抑制其活性,呈現雙向作用,文獻中未見報導,在本研究首見。以多巴色素自動氧化生成黑色素量試驗中,在濃度500µg/ml時,以積雪草具較明顯抑制作用(42±0.3%)。另外,在擬超氧化物歧化酶活性試驗中,以濃度0.05g測試時,則以絲瓜葉、台灣木通、積雪草具較明顯之抗氧化能力。 綜合以上結果,顯示15種中草藥之乙醇萃取物中,以積雪草同時具較明顯之抑制黑色素生合成活性與抗氧化能力,然其活性成分及相關機轉仍有待進一步探討。; Abstract Alcoholic extracts of fifteen kinds of Chinese herbs ( Centella asiatica (L.) Urban, Rabdosia lasiocarpus (Hayata) Hara, Patrinia scabiosaefilia Fisch, Sophora tonkinensis Gapnep, Akebia longeraecemosa Matsum, Duchesnea chrysantha (Zucc.&Mor.)Miq, Polygonum plebeium R. Br, Saxifraga stolonifera Meerb, Myrica rubra Sieb. & Zucc. var. acuminata Nakai, Perilla ocymoides L. var. crispa Benth., Forma purpurea Makino, Luffa cylindrica (L.) M. Roem, Siegesbeckia glabrescens Makino, Areca catechu L., Piper nigrum L., and Piper betle L., )were assayed in vitro for their inhibition of melanogenesis and antioxidant capacity, individually. Among them, the results demonstrated that the extract of Saxifraga stolonifera Meerb exerted significantly inhibitory effect(42.9±0.5%) on tyrosinase activity at lower concentration(50µg/ml), the extract of Centella asiatica (L.) Urban exerted significantly inhibitory effect(44.9±0.4%)at moderate concentration (200µg/ml), and the extract of Piper nigrum L. exerted significantly inhibitory effect(41.0±0.5%)at higher concentration (500µg/ml). However, the extract of Rabdosia lasiocarpus (Hayata) Hara showed inhibition at lower and doses higher, activation at moderate dose,. The biphasic inhibition was found first time in this study. The extract of Centella asiatica (L.) Urban exerted significantly inhibitory effect(42±0.3%)on dopa production of melanin from dopachrome by autooxidation at 500µg/ml. In addition, the extract of Luffa cylindrica (L.), Akebia longeraecemosa Matsum, and Centella asiatica (L.) Urban exerted significant antioxidation on SOD-like test at 0.05g. In conclusion, only the extract of Centella asiatica (L.) Urban showed both significant effects on inhibition of melanogenesis and antioxidant capacity. Thus, these results provided important information for further investigation, although the active constituents and mechanism are unknown.
