    題名: 延胡索、益母草及雞血藤對高脂血大鼠降血脂作用之研究;Studies on the hypolipidemic effect of Corydalis Rhizoma, Leonuri Herba and Spatholobi Caulis in hyperlipidemic rats

    摘要: 本研究探討活血化瘀藥益母草、化瘀止痛藥延胡索及養血活血藥雞血藤之甲醇粗抽物與延胡索成分四氫巴馬汀(THP)對高膽固醇食物誘導高脂血症大鼠降血脂作用之影響。首先以高膽固醇食物(膽固醇2 %及膽酸1 %)每天口服一次持續4天誘導產生大鼠高脂血,第四天起同時再口服給予活血化瘀藥治療。於第7、10、14天檢測其血清、肝臟組織及糞便之血脂生化值,以探討其調節血中脂質之作用及其可能機轉。 實驗結果顯示延胡索甲醇粗抽物對高脂血症大鼠之血清總膽固醇、低密度脂蛋白及肝臟之三酸甘油酯皆有顯著降低,血清脂蛋白脂酶則明顯提升,肝臟之總膽固醇與糞便之膽固醇及膽酸無顯著影響;益母草甲醇粗抽物對高脂血症大鼠之血清總膽固醇、低密度脂蛋白及肝臟之三酸甘油酯亦有明顯降低,且高密度脂蛋白及脂蛋白脂酶顯著增加,肝臟之總膽固醇與糞便之總膽固醇及膽酸無顯著影響。雞血藤甲醇粗抽物對高脂血症大鼠不具降血脂之作用。另外四氫巴馬汀對高脂血症大鼠之血清總膽固醇、低密度脂蛋白及肝臟之三酸甘油酯顯著降低,血清高密度脂蛋白則顯著提高,脂蛋白脂酶、肝臟之總膽固醇及糞便之膽固醇與膽酸則無顯著影響。 綜合以上結果顯示,延胡索之甲醇粗抽物可經由提高血清脂蛋白脂酶活性以加速血中脂質代謝,達到降血脂作用;益母草可經由提高脂蛋白脂酶活性以加速血中脂質代謝及提高血清高密度脂蛋白濃度以加速血中膽固醇清除,達到降血脂作用;THP可經由提高血清高密度脂蛋白濃度以加速血中膽固醇清除,達到降血脂作用。; The purpose of this study was to investigate the hypolipidemic effect of methanol extract of drugs such as Leonurus heterophyllus which is used to promoting blood circulation and remove blood stasis, Corydalis Rhizoma used to remove blood stasis and arrest pain, and Spatholobi Caulisin used to nourishing blood and promoting blood circulation, and dl-tetrahydropalmatine (THP) in the hyperlipidemic (HYP) rats induced by high cholesterol diet. Firstly, high cholesterol diet (cholesterol 2 % and bile acid 1 %) were administered orally to rats once a day for four days could induce hyperlipidemia. The above mention drugs were treated orally with from the fourth day. To probe into the regulation function of blood lipid and its possible mechanism, the lipid biochemical values of serum, liver tissue and fecal were measured at the 7th, 10th, and 14th day. The results shown that CR apparently reduced total cholesterol (TC) of serum, low-density lipid (LDL) of serum and triglyceride (TG) of the liver. Furthermore, it obviously increased lipoprotein lipase (LPL) activity. However, it did not significantly affect TC of the liver, TC and bile acid (BA) of the fecal in the HYP rats. LH also obviously reduced TC of serum, LDL of serum and TG of the liver. It significantly increased HDL and LPL of serum. Besides, it did not significantly affect TC of the liver, TC and BA of the fecal on the HYP rats. SC did not apparently affect the lipid biochemical values of serum, liver tissue and fecal in the HYP rats. THP obviously reduced TC and LDL of serum, and TG of the liver. In addition, it apparently increased HDL of serum, and did not apparently influence LPL of serum, TC of the liver, and TC and BA of fecal in the HYP rats. The above results demonstrated that methanol extract of CR have hypolipidemic effect by increasing LPL activity of serum to accelerate metabolism of blood lipid. Methanol extract of LH have hypolipidemic effect by increasing LPL activity of serum to accelerate metabolism of blood lipid and by serum level of HDL to remove blood cholesterol rapidly. Moreover, THP has hypolipidemic effect by increasing serum level of HDL of serum to remove blood cholesterol rapidly.
