摘要: | 榼藤(Entada phaseoloides (LINNE) MERRILL)之化學成分研究 陳兆群 中國醫藥學院 中國藥學研究所 摘要 台灣產藥用植物榼藤(Entada phaseoloides (L.) MERR.)為豆科(Leguminosae)植物,又名鴨腱藤、大血藤、過崗龍等別名。廣泛分部於台灣中部山區原始闊葉林、再生林或雨林地內。具有袪風除濕,活血通絡,通經止痛,消腫解毒之效。治四肢麻木,風濕腰痛,風濕性關節炎,跌打損傷,癰瘡腫毒,為固有之台灣民間藥,其毒性經本研究測得其莖部甲醇抽取物之LD50為170 mg/kg,在藥理活性方面,發現氯仿層、乙酸乙酯層、正丁醇層等對HONE-1(人類鼻咽癌細胞)有抑制活性,而乙酸乙酯層和正丁醇層等對NUGC(人類鼻胃癌細胞)也有抑制活性。而榼藤甲醇粗抽物,在DPPH自由基清除能力之試驗發現正丁醇層、P層、甲醇粗抽等具有明顯之活性。因民間常用來治療風濕腰痛及風濕性關節炎等,故本研究亦進行其鎮痛、抗炎之藥理活性評估,已確認其莖部甲醇粗抽物在5mg/kg時,確實有明顯的抗炎及鎮痛作用。 自榼藤莖部之甲醇出抽物分離出的成分有spinasterol (H-1)、5, 4’-dihydroxy-3, 7, 3’-trimethoxyflavone (CL-2)、triacontanoic acid (CL-3)、spinasterol-b-D- glucoside (CL-4)、quercetin (EA-2)以及EA-S (仍在解析中)等成分,而上述化合物均為第一次由本植物中分離得的成分。 綜合以上結果可知,榼藤莖部有明顯的細胞毒殺、抗氧化、抗炎與鎮痛等作用。而由本實驗所分離出成分之藥理活性,則有待更進一步之探討與確認。; Studies on the Chemical Constituents of Entada phaseoloides (LINNE) MERRILL Jaw-Chyun Chen Institute of Chinese Pharmaceutical Sciences China Medical College Abstract Entada phaseoloides (L.) MERR. is a native plant of Taiwan belonging to the Leguminosae family. It is a common folk medicine being used for tetraplegia, rehumatalgia, traumatic injury and ulcerative carbuncle. Our preliminary test showed that the CHCl3, EtOAc and n-BuOH fractions of E. phaseoloides exhibited significant cytotoxic activity against NUGC & HONE-1 cell lines. We also evaluated the DPPH free radical scavenging, anti-inflammatory and analgesic effects of the methanol extract of the stem of Entada phaseoloides. The LD50 of methanol extract was 170 mg/kg. The methanol extract showed significant anti-inflammation actions by inhibiting rat paw swelling. It also showed analgesisic effect by decreasing the frequencies of leg-licking in formalin induced pain in mice. In DPPH free radical scavenge screening test, the methanol extract, n-BuOH fraction and precipitate fraction showed significant antioxidant activity. From its methanolic extracts, six compounds were isolated : spinastero l(H-1), 5, 4’-dihydroxy-3, 7, 3’-trimethoxyflavone (CL-2), triacontanoic acid (CL-3), spinasterol-b-D-glucoside (CL-4), quercetin (EA-2) and compound EA-S. These compounds were first isolated from this plant and their structures were determined by spectroscopic methods. Our result indicated that extract Entada phaseoloides exhibted significant cytotoxic activity, antioxidant activity, analgesisic effect and anti-inflammatory effect. However, the active constituents responsible for the activities are still under investigation. |
