    摘要: 由豆科植物泰葛(Pueraria Mirifica) 醇抽物中,共分離而得四個屬於植物雌激素的化合物:一個為雜環多酚類的葛雌素(+)-Miroestrol (I),三個為異黃酮骨架的葛根素(Puerarin) (Ⅱ)、大豆?元(Daidzein) (Ⅲ)及染料木黃酮(Genistein) (Ⅳ)。前述化合物(I)~ (Ⅳ)加上大豆素(Daidzin) (Ⅴ)及染料木?(Genistin) (Ⅵ)對HL-60細胞株的抗增殖活性,顯示除葛根素(Ⅱ)外,其他活性皆很好。(+)-Miroestrol (I)為首次測試HL-60血癌細胞抗增殖活性,IC50 = 0.11mM。 泰葛(PM)、大陸四川產及台灣產葛根共七個樣品,利用HPLC系統定量葛根素(Ⅱ) 、大豆素 (Ⅴ)、大豆?元 (Ⅲ)、染料木黃酮 (Ⅳ)及染料木?(Ⅵ)等五個活性成分的含量。泰葛原植物(PM)葛根素最高,台灣產葛根(PP及PT)的大豆素(Ⅴ)、大豆?元 (Ⅲ)、染料木?(Ⅵ)等含量為其中最高者。 泰葛一年生根、莖和葉及由種子、根和莖誘發愈傷組織的活性成分含量也經由HPLC定量出來。在大豆?元 (Ⅲ)含量上,以種子之愈傷組織含量最高。在染料木黃酮 (Ⅳ) 含量上,以根之愈傷組織含量最高。 葛雌素(I)在泰葛成分上的定量HPLC系統,首次被建立。結果顯示確實為泰葛特有成分。; Four phytoestrogens ─ A heterocyclic phytoestrogen, (+)-Miroestrol (I) and three isoflavones, puerarin (II), daidzein (III) and genistein (IV) , have been isolated from alcoholic extracts of dried roots of Pueraria mirifica. Compound (I) ~ (IV), daidzin (V) and genistin (VI) were screened for their antiproliferative activities of HL-60 Cells. Besides Puerarin(II), all compounds showed good abilities of antiproliferation. (+)-Miroestrol (I) was the first time to evaluate antiproliferative effect and Its IC50 was 0.11mM. By using HPLC system, the contents of puerarin (II), daidzein (III), genistein (IV), daidzin (V) and genistin (VI) in dried roots of Pueraria mirifica (PM) and six samples of Radixs Pueraria from Szechuan in Mainland and Taiwan were assayed. Dried roots of PM had the highest contents of puerarin (II) and contents of daidzein (III), Genistein (IV) Daidzin (V) and Genistin (VI) were higher in Taiwan’s “Ge-Ken”. The contents of Compound (II)~ (VI) in roots, stems and leaves which were one years old and in calli induced from seeds, roots and stems were measured,too. The callus from seeds had more dadzein and genistein was higher amounts in roots of the callus. The HPLC system to assay (+)-Miroestrol (I) was the first time to be established and there are no (+)-Miroestrol (I) in other Radixs Puerariae.
