    摘要: 中文摘要 本研究之目的在探討生甘草對關木通所導致的急性腎毒性的保護作用,研究採用口服餵食給藥的模式,選取48隻SD雌性大白鼠,隨機分組成對照組、關木通(30 g/Kg)組、30 g/Kg 關木通加1 g/Kg 生甘草組、關木通(20 g/Kg)組、20 g/Kg 關木通加1 g/Kg 生甘草組、 關木通(10 g/Kg)組、10 g/Kg 關木通加1 g/Kg 生甘草組、生甘草1 g/Kg 組。經一週的餵食之後,記錄其實驗前後的體重、進食量、飲水量、尿量、活動力的變化以及其腎臟組織病理變化。檢驗大鼠尿中Protein的含量,血中BUN, Creatinine的數值。結果分別以不同劑量的關木通組互相比較,另外關木通組再分別與同劑量之關木通配伍生甘草組以Fisher’s exact test及Kruskal-Wallis test進行分析,並以Duncan’s Multiple-Range Test作事後檢定,之後再使用Two factor of ANOVA 來探討兩者之間的交互作用。實驗後的研究結果顯示,隨著關木通之劑量升高,血中BUN, Creatinine的數值隨之上升,活動力、進食量、體重增加量隨之減少,腎臟病理病變增大。關木通組與同劑量之關木通配伍生甘草組相比較,血中BUN, Creatinine的數值上升較多,活動力、進食量、體重增加量減少,腎臟病理病變較大。顯示關木通之劑量愈大,對腎臟的急性傷害愈大。並証實了生甘草可減輕關木通的急性腎臟傷害以及胃部傷害。其機轉可能與甘草的皮質樣激素有關,但詳細機轉為何,仍需進一步探討。; The Effect of Gancao on Guamutong-induced Acute Renal Toxicity in Rats Li-Hsiang Wang Institute of Chinese Medical Science, China Medical College Abstract The purpose of this experiment was to investigate how the effect of Gancao on Guamutong-induced acute renal toxicity in rats. Forty-eight female SD rats were randomly separated into eight groups – a control group; a group of 30 g/Kg of Guamutong in rat; a group of 30 g/Kg of Guamutong combined with 1 g/Kg of Gancao in rat; a group of 20 g/Kg of Guamutong in rat; a group of 20 g/Kg of Guamutong combined with 1 g/Kg of Gancao in rat; a group of 10 g/Kg of Guamutong in rat; a group of 10 g/Kg of Guamutong combined with 1 g/Kg of Gancao in rat; a group of 1 g/Kg of Gancao in rat. Rats in the eight groups were given different doses of drugs for one week using the gastric tube. We surveyed the general condition, the histology of the kidney, the data of the renal function of blood and urine in the rats. Then we analyzed the results using the SAS packages. Between the groups with different doses of the Guamutong, we found the rats’ weight and activity dropped significantly and necrosis area of their epithelium in the renal tubules as well as the biochemical level of BUN and creatinine rose significantly, and these results were dose-dependent. However, these effects of Guamutong were attenuated as feeding the rats with the Gancao. These results indicated that the Gancao can decrease the acute renal toxicity of Guamotong based on the histological and laboratory observations.
