摘要: | 胎盤素的療效已在民俗療法中得到很多佳評,但其藥理機制所知有限且眾說紛紜,而應用在外傷的傷口癒合的藥理分子機制更有待進一步去探討證實? 胎盤粹取物中含有蛋白質月生月太 類生長因子, 可經由細胞組織間的autocrine或paracrine的作用機制,對於促進皮膚的修復再生方面應具有重要的角色? 本篇論文之研究結果顯示, 藉由體內(in vivo) 和體外(in vitro) 給予豬胎盤粹取物後,纖維母細胞(NIH 3T3 cells,mice embryo fibroblast cells)的增生作用可被明顯的促進? 利用西方轉漬法(Western blotting analysis)所得到之結果顯示, 給予豬胎盤粹取物後的纖維母細胞之細胞培養和燙傷處理的老鼠皮膚中,可增加纖維母細胞生長因子(fibroblast growth factor,FGF)及細胞局部趨化激酉每(focal adhesion kinase,FAK)之蛋白質表現量 ; 而利用流式細胞分析儀(flow cytometry analysis)分析細胞生長週期中細胞分佈在S-phase的百分比,結果顯示以每毫升含有1、10、30毫克胎盤粹取物等三個劑量來培養纖維母細胞, 其S-phase 所增加的百分比相較於對照組, 分別增加有二倍、三倍及四倍之多。在燙傷的老鼠皮膚試驗中, 分別給予每毫升含有1、10、100毫克等三個劑量的豬胎盤粹取物,並於1-16天連續給藥處理的燙傷處,每天取一生檢樣品(biopsy sample)作組織病理學切片並加以分析。根據我們的研究結果發現, 利用豬胎盤粹取物所處理的實驗組,其傷口癒合的天數與對照組比較具有一明顯的統計意義存在? 所以,胎盤粹取物的藥理機制, 可由本實驗對於傷口癒合的促進結果, 得到一有力的科學性之數量化的佐證 ?; The pharmacological molecular mechanisms of placenta on wound healing are not fully understood. However, its constitutents of peptide growth factors acting through autocrine or paracrine mechanism were expected to play an important role. Our in vivo and in vitro studies have demonstrated that the topical application of placenta extract accelerated the rate of fibroblast cell proliferation. Western blotting analysis of the NIH 3T3 cells revealed an increased levels of fibroblast growth factor (FGF) and focal adhesion kinase (FAK) under the effects of placenta extracts. To demonstrate the fortification effects of placenta through the process of wound healing, cultured NIH 3T3 cells treated with placenta extract were subjected to flow cytometric analysis. There was a 4 folds, 3 folds, 2 folds increase in 30 mg/ml, 10 mg/ml, 1 mg/ml placenta-treated group as compared to the controls. In the in vivo study, rats were burned on the back and treated with 100 mg/ml, 10 mg/ml, 1 mg/ml of placenta extracts. These rats were sacrificed 16 days after burn and their skins were harvested for histopathological analysis. A significant increase in scar area of wound healing was found in the porcine placenta extract-treated group as compared to the controls. These results suggest that placenta extract may be a potential pharmacological reagent in skin wound healing. |
