    摘要: 由台灣竹山土壤中篩選所得之竹山鏈黴菌 (Streptomyces chusanensis, ZS-2),產生一新的第II型核酸限制,命名為SchI。純化後的SchI,經鑑定為一具有次單元分子量28kDa的同雙聚體,且無外切之活性。此一限制切磷二酯鍵,所切的位置在DNA順序5'- CCGCGG-3’的第四個位置上的C與第五位置上的G之間,為目前已被廣泛應用的SacII的同切。在不同的反應條件來比較SchI與SacII,發現SchI較SacII更適合用於各種基因基礎和應用的研究。

    Streptomyces chusanensis ZS-2, isolated from a soil sample in Chusan of Taiwan, was found to produce a new Type II restriction endonuclease. This restriction enzyme was designated as SchI. The purified enzyme was characterized to be a homodimer with a subunit molecular weight of 28 kDa. It was apparently free from exonuclease activities. It cleaves the phosphodiester bond between the 4th "C" and the 5th "G" on the 5'- CCGCGG-3' sequence of DNAs leaving a 2-nucleotide protruding end at its 3' site. This data suggests that SchI is an isoschizomer of SacII. In addition, based on the comparison between SchI and SacII regarding of reaction parameters, it seems that SchI is a better choice of restriction enzyme for genetic analysis and mapping.
