    Title: 中草藥抑制黑色素生合成之初步評估研究 (論文編號: P2-12)

    Abstract: 本研究分別以酪胺酸?活性試驗,多巴色素自動氧化生成黑色素量試驗和擬超氧化物歧化?活性試驗,評估15種中草藥:積雪草(Centella asiatica Urban)、毛果延命草(Rabdosia lasiocarpus Hara)、黃花敗醬草(Patrinia scabiosaefilia Fisch)、山豆根(Sophora tonkinensis Gapnep)、台灣木通(Akebia longeracemosa Matsum)、台灣蛇莓(Duchesnea chrysantha Miq.)、小萹蓄(Polygonum plebeium R. Br.)、虎耳草(Saxifraga stolonifera Meerb)、銳葉楊梅(Myrica rubra Sieb. & Zucc. var. acuminate Nakai)、紫蘇葉(Perilla ocymoides L. var. crispa Benth. forma purpurea Makino)、絲瓜葉(Luffa cylindrica M. Roem)、毛梗豨薟草(Siegesbeckia glabrescens Makino)、檳榔(Areca catechu L.)、胡椒(Piper nigrum L.)及荖葉(Piper betel L.)之乙醇萃取物對抑制黑色素生合成與抗氧化之活性。體外試驗結果顯示,在對酪胺酸?活性試驗中,低濃度(50?g/ml)時,以虎耳草對酪胺酸?具較明顯的抑制作用(42.9±0.5%);中濃度(200?g/ml)時,以積雪草對酪胺酸?較具明顯的抑制作用(44.9±0.4%);高濃度(500?g/ml)時,以胡椒對酪胺酸?具較明顯的抑制活性(41.0±0.5%)。然在不同濃度測試中,毛果延命草於低濃度(50?g/ml)時則對酪胺酸?具抑制活性,中濃度(200?g/ml)時具促進活性,高濃度(500?g/ml)時對酪胺酸?則具抑制活性,而呈現雙向作用,文獻中未見報導,在本研究為首見。以多巴色素自動氧化生成黑色素量試驗中,在濃度500?g/ml時,以積雪草具較明顯抑制作用(42±0.3%)。另外,在擬超氧歧化?活性試驗中,以濃度0.05g測試時,則以絲瓜葉、台灣木通、積雪草具較明顯之抗氧化能力。綜合以上結果,顯示15種中草藥之乙醇萃取物中,以積雪草同時具較明顯之抑制黑色素生合成活性與抗氧化能力,然其活性成分及相關機轉仍有待進一步探討。 關鍵字:中草藥(Chinese herbs),酪胺酸?(tyrosinase),黑色素(melanin),多巴色素(dopachrome),擬超氧歧化?活性試驗(SOD-like test)
