    題名: Phylogenetic analysis of the internal transcribed spacers of ribosomal DNA of Corchorus in Taiwan?

    摘要: The genus Corchorus (Tiliaceae) has 4 species, C. capsularis, C. olitorius, C. aestuans and C. aestuans var. brevicaulis. A delicious soup called Ma-Yih in central Taiwan is actually made of C. capsularis or C. olitorius which is traditionally deemed has anti-inflammatory and antipyretic effects. They are introduced and promoted by Farmers’ Association in Taichung as health foods. They are various but resemble in morphology. For developing and using these resources, this study is carried out to identify the gene sequence on Corchorus. The entire internal transcribed spacer (ITS) region between 18S and 28S ribosomal RNA of the four Corchorus species was amplified by polymerase chain reaction (PCR). The primers for ITS1 is 18D (5’-CACACCGCCCGTCGCTCCTACC GA-3’) and 1858 (5’-TTGCGTTCAAAGACTCGATG-3’), ITS2 is 2858 (5’-CAACG GATATCTCGGCTCTC-3’) and 28CC (5’-ACTCGCCGTTACTAGGTGAA-3’), that designed for amplification and sequencing. The length of PCR product was 853~864 bp. Comparing the obtained sequence with the sequence of ITS from other higher plant species it showed that the cloned sequence contained 70 bp of 28S rRNA partial sequence, ITS region and 5.8S complete sequence, and 120 bp of 18S rRNA partial sequence. The length of ITS region was 663~674 bp in Corchorus species, including 275~279 bp of ITS1, 162 bp of 5.8S rRNA, and 223~233 bp of ITS2. The G+C contents of ITS1, 5.8S and ITS2 regions were 59.7~63.1%, 54.3~58% and 60.5~62.7% respectively. In this study, the sequence similarity of the rRNA region between Corchorus species was 92~99%. It showed that four gene sequences of Corchorus species are very close. It can provide a clear and definite evidence to identify Corchorus species.?
