    Title: KC-2T美白作用安全性及其機轉探討(2/2)

    Abstract: 於前年度研究計畫中具美白作用之植物成分,發現KC-2T(100 μg╱mL) 在黑色素生成試驗其抑制效果較熊果甘(2.72 mg╱mL)佳。KC-2T是由茜草科植物九節木經適當萃取及處裡而得。本計畫就KC-2T安全性評估進行細胞毒性,眼黏膜刺激及皮膚一次性試驗之探討,結果顯示KC-2T 無細胞毒性,對眼黏膜有輕微刺激性,而皮膚一次性試驗則無皮膚刺激性。第一年的計畫亦發現九節木成分中的vanillic acid (16.8 mg╱mL)於平版試驗中較熊果甘(27.2 mg╱mL)佳之抑制效果。所以本計畫就vanillic acid的美白作用進行機轉探討。結果顯示Vanillic acid於黑色素生成作用機轉中黑色素生成及抑制酪胺酸甘合成試驗中並無顯著的抑制作用,然而在直接阻斷小鼠黑色素瘤細胞酪胺酸甘活性則有抑制效果。

    Our previous study on whitening constituents-containing plants showed that the inhibition of melanin synthesis for KC-2T (100 ug╱mL) was stronger than that of arbutin (2.72 mg╱mL). KC-2T was derived from Psychotria rubra (LOUR.) POIR. of Rubiaceae after suitable extraction and treatment. In this project, the safety of KC-2T cytotoxicity, the eye irritation test and primary skin irritation test were carried out. The results showed that KC-2T was non-cytotoxicity, mild irritation and non-skin irritation. In the first year project, vanillic acid (16.8 mg╱mL), the constituent in the plant containing Psychotria rubra (LOUR.) POIR., showed stronger inhibition of tyrosinase activity by means of inhibition zone test than that of arbutin (27.2 mg╱mL). In this project, the mechanisms of whitening effect of vanillic acid were studied. The result showed that vanillic acid had no good effect on the synthesis of tyrosinase. However, vanillic acid had inhibitory effect on tyrosinase activity in B16 mouse cell.
