摘要: | 苦瓜(Momordica Charantia)在亞洲地區是一種日常食用蔬菜,由於被發現含有活化PPARα及PPARγ成分,因此具有高度潛力可研發為對抗代謝症候群保健食品。本項子計劃目標在研究苦瓜之抗體脂堆積功能及成分,擬分三年進行,第一年(93年)我們已成功利用in vivo模式證明苦瓜凍乾粉如同治療糖尿病藥物Thiazolidinedione可減輕高脂高糖飲食誘發大鼠之高胰島素血症,卻沒有肥胖副作用,5%苦瓜凍乾粉劑量可有效抑制腹部脂肪細胞肥大。第二年擬以In vitro 模式篩選苦瓜降體脂功能成分,計劃以前脂肪細胞株3T3-L1 及初代培養分離自大鼠脂肪組織之stroma-vascular cell(SV cells),在不同階段處理不同品種苦瓜乙酸乙酯(EA)萃取物,探討:(1)苦瓜萃取物對前脂肪細胞增生(Proliferation)影響;(2) 對前脂肪細胞分化(differentiation)為脂肪細胞影響;(3)對已分化脂肪細胞去脂(delipidation)影響。目前結果顯示五種苦瓜(110310、2381、8153、HM18 及K922)EA萃對3T3-L1細胞作用,以2381 有最強的促增生與最弱的促分化作用,而8153則有最弱的促增生與最強的促分化作用,是否暗示2381 有最好的降體脂功能有待研究;此外定量苦瓜萃中conjugated linolenic acid (CLN)方法及CLN對脂肪細胞分化影響之研究進行中。
Recently, Bitter Melon (Momordica charantia; BM) was found to activate both peroxisome proliferator-activated receptor (PPAR)αandγwhich may be valuable in prevention and amelioration metabolic syndrome. This three-year study was aimed at the anti-adiposity effect of BM, and tried to find out the functional components. In the first year, we had successfully proven that BM powder, as well as Thiazolidinedione (TZD), ameliorate hyperinsulinemia induced by high fat diet in rats. However, there was no obesity side effect of TZD was shown in BM-fed rats. BM powder was effective in inhibiting the hypertrophy of abdominal adipocytes at 5% dietary level. In the second year, an in vitro medel for screening the functional components with anti-adiposity effect of BM will be established. We used a preadipocyte cell line, 3T3-L1, and primary cell culture, stroma-vascular cell (SV cells) isolated from adipose tissue of rats, treated with EA extract of different species of BM at different cell stage. To evaluate: (1) effect of BM extract on proliferation of preadipocytes. (2) effect of BM extract on differentiation of preadipocytes. (3) effect of BM extract on delipidation of differentiated adipocytes. Until now, our study showed 2381 was the most potent specie of BM in enhancing preadipocyte proliferation, and the weakest in enhancing adipocyte differentiation. In contrary, 8153 had the weakest effect in promoting cell proliferation but was the most potent in enhancing cell differentiation. In addition, the work of quantification of conjugated linolenic acid (CLN) in EA extract of five species of BM, and the effect of CLN on adipocyte differentiation was in progress. |
